胃癌细胞上清液对腹膜间皮细胞的作用
         
Effect of supernatant derived from gastric cancer cells on peritoneal mesothelial cells

摘    要
目的: 探讨胃癌细胞上清液对腹膜间皮细胞的作用。方法: 用胃癌细胞上清液处理人腹膜间皮细胞后,通过HE染色法观察腹膜间皮细胞的形态学变化,免疫细胞化学法检测腹膜间皮细胞中上皮细胞标志蛋白E-cadherin、cytokeratin和间质化标志蛋白N-cadherin、fibronectin的表达。将胃癌细胞上清液注射入裸鼠腹腔,应用免疫组织化学法和蛋白质印迹法检测裸鼠腹膜组织中上皮细胞标志蛋白E-cadherin、cytokeratin和间质化标志蛋白N-cadherin、fibronectin的表达。结果: 经胃癌细胞上清液处理的腹膜间皮细胞呈梭形,细胞脱落,细胞间连接减少;胃癌细胞上清液处理的腹膜间皮细胞E-cadherin和cytokeratin的表达水平低于未处理的空白对照组和用无血清DMEM培养液处理的实验对照组(P值均<0.05),而N-cadherin和fibronectin的表达水平则高于未处理的空白对照组和用无血清DMEM培养液处理的实验对照组(P值均<0.05)。腹腔注射胃癌细胞上清液的裸鼠腹膜组织E-cadherin和cytokeratin的表达水平低于未处理的空白对照组和腹腔注射无血清DMEM培养液的实验对照组(P值均<0.05),而N-cadherin和fibronectin的表达水平则高于未处理的空白对照组和腹腔注射无血清DMEM培养液的实验对照组(P值均<0.05)。结论: 胃癌细胞上清液可使腹膜间皮细胞失去原有的上皮细胞生物学行为,而获得间质细胞的生物学行为。

Abstract
Objective: To investigate the effect of supernatant derived from gastric cancer cells on peritoneal mesothelial cells. Methods: The human peritoneal mesothelial cells were treated with supernatant derived from gastric cancer cells, then the change of morphology of peritoneal mesothelial cells was observed by using hematoxylin-eosin (HE) staining; the expressions of protein markers E-cadherin and cytokeratin for epithelial cells and the protein markers N-cadherin and fibronectin for mesenchymal cells were detected by immunocytochemistry. The supernatant derived from gastric cancer cells was injected into abdominal cavity of nude mice, then the expressions of E-cadherin, cytokeratin, N-cadherin and fibronectin in peritoneal tissues in nude mice were detected by immunohistochemistry and Western blotting, respectively. Results: After treatment with the supernatant derived from gastric cancer cells, the peritoneal mesothelial cells looked spindle-shaped and dropped, with weakened intercellular adhesion. The expression levels of E-cadherin and cytokeratin in peritoneal mesothelial cells after treatment with supernatant derived from gastric cancer cells were lower than those in the peritoneal mesothelial cells without any treatment (as blank control) or cultured with serum-free DMEM medium (as experimental control) (all P<0.05), while the expression levels of N-cadherin and fibronectin were higher (all P<0.05). The expression levels of E-cadherin and cytokeratin in peritoneal tissues in nude mice after intraperitoneal injection with supernatant derived from gastric cancer cells were lower than those in peritoneal tissues in nude mice without any treatment or intraperitoneally injected with serum-free DMEM medium (all P<0.05), while the expression levels of N-cadherin and fibronectin were higher (all P<0.05). Conclusion: The peritoneal mesothelial cells may lose biological behaviors of epithelial cells, meanwhile getting the biological behaviors of peritoneal mesothelial cells via intervention with supernatant derived from gastric cancer cells.
