葛根素对激素非依赖性前列腺癌细胞增殖、凋亡、迁移及侵袭的作用
         
Effects of puerarin on proliferation, apoptosis, migration and invasion of hormone-independent prostate cancer cells

摘    要
目的:探讨葛根素对激素非依赖性前列腺癌细胞增殖、凋亡及迁移的影响及其可能的作用机制。 方法:应用CCK-8法和FCM法分别检测葛根素对前列腺癌PC3细胞增殖、凋亡和细胞周期的影响,应用Transwell小室法检测葛根素对前列腺癌PC3细胞迁移和侵袭的影响,应用实时荧光定量PCR法和蛋白质印迹法分别检测葛根素对前列腺癌PC3细胞中凋亡相关基因和蛋白表达及蛋白激酶B(protein kinase B,PKB,又称Akt)活性的影响。 结果:与未干预的对照组相比,不同浓度的葛根素可显著抑制PC3细胞的增殖(P值均<0.05),诱导其凋亡(P值均<0.01),并可将PC3细胞阻滞于G2/M期(P值均<0.05),亦可有效抑制PC3细胞的迁移和侵袭(P值均<0.01)。不同浓度葛根素处理组PC3细胞中磷酸化Akt(phospho-Akt,p-Akt)蛋白(P值均<0.01)及抗凋亡分子Bcl-2(B-cell lymphoma-2)mRNA(P值均<0.05)和蛋白(P值均<0.01)的表达水平均明显低于对照组,促凋亡分子Bax(Bcl-2-associated X)mRNA(P值均<0.05)和蛋白(P值均<0.01)、Fas(factor associated suicide)mRNA(P值均<0.01)、caspase-3 mRNA(P值均<0.01)、cleaved caspase-3蛋白(P值均<0.01)及caspase-8 mRNA(P值均<0.01)的表达水平均高于对照组。 结论:葛根素具有抑制激素非依赖性前列腺细胞增殖、迁移及侵袭的作用,并可通过上调caspase-3的活性,促进细胞凋亡。

Abstract
Objective: To investigate the effects of puerarin on the proliferation, apoptosis and migration of hormone-independent prostate cancer cells, and to explore the possible mechanisms. Methods: The effects of puerarin on the proliferation, apoptosis and cell cycle distribution of prostate cancer PC3 cells were detected by CCK-8 and FCM assay, respectively. The effects of puerarin on the migration and invasion of prostate cancer PC3 cells were detected by Transwell chamber assay. The effects of puerarin on the expressions of apoptosis-associated genes and proteins as well as the activity of protein kinase B (PKB, also known as Akt) in prostate cancer PC3 cells were detected by real-time fluorescent quantitative PCR and Western blotting, respectively. Results: As compared with the control group without intervention, the different concentrations of puerarin could significantly inhibit the proliferation of PC3 cells (all P < 0.05), induce the apoptosis of PC3 cells (all P < 0.01), arrest the PC3 cells at G2/M phase (all P < 0.05), and also effectively inhibit the migration and invasion of PC3 cells (all P < 0.01). The expression levels of phospho-Akt (p-Akt) protein (all P < 0.01) and anti-apoptotic molecule B-cell lymphoma-2 (Bcl-2) mRNA and protein (all P < 0.05) in PC3 cells treated with different concentrations of puerarin were significantly lower than those in the control group. The expression levels of apoptotic molecule Bcl-2-associated X (Bax) mRNA (all P < 0.05) and protein (all P < 0.01), factor associated suicide (Fas) mRNA (all P < 0.01), caspase-3 mRNA (all P < 0.01), cleaved caspase-3 protein (all P < 0.01) and caspase-8 mRNA (all P < 0.01) in PC3 cells treated with different concentrations of puerarin were significantly higher than those in the control group. Conclusion: Puerarin can inhibit the proliferation, migration and invasion of hormone-independent prostate cancer cells, and also promote apoptosis by up-regulating the activity of caspase-3.
