T细胞免疫球蛋白黏蛋白3在人食管鳞癌组织中的表达及其临床意义
         
Expression of T-cell immunoglobulin-and mucin domain-containing protein-3 in human esophageal squamous cell cancer tissues and its clinical significance

摘    要
目的:检测T免疫球蛋白黏蛋白3(T-cell immunoglobulin-and mucin domain-containing protein-3,TIM-3)在食管鳞癌组织中的表达情况,并分析TIM-3表达与患者临床理特征及术后生存时间的关系。
方法:采用实时荧光定量PCR法和免疫组织学法分别检测食管鳞癌组织及其相应的癌旁正常组织中TIM-3 mRNA及蛋白的表达水平。采用χ2检验及Fisher精确概率法分析TIM-3蛋白表达与食管鳞癌患者临床理参数之间的关系。采用Kaplan-Meier分析法比较不TIM-3蛋白表达水平对患者术后生存时间的影响。
结果:食管鳞癌组织中TIM-3 mRNA的表达水平较癌旁组织显著升高(P<0.001)。TIM-3蛋白在食管鳞癌组织中的阳性表达率为85.9%(55/64),而其在癌旁正常组织中表达很弱或几乎不表达,差异有统计学意义(P<0.001)。食管鳞癌组织中TIM-3蛋白的表达与浸润深度、淋巴结转移及TNM分期密切相关(P值均<0.05)。食管鳞癌组织中TIM-3蛋白的表达水平与患者预后呈负相关性(P<0.05)。
结论:TIM-3异常表达可能与食管鳞癌的发生和发展密切相关,提示TIM-3在食管癌的早期诊断、预后判断及靶向治疗中可能具有潜在的临床应用价值。

Abstract
Objective:To detect the expression of T-cell immunoglobulin-and mucin domain-containing protein-3(TIM-3) in human esophageal squamous cell carcinoma(ESCC) tissues, and to evaluate the correlation between TIM-3 expression and clinicopathological features and postoperative survival of patients with ESCC.
Methods:The expressions of TIM-3 mRNA and protein in ESCC tissues and the corresponding para-cancerous normal tissues were detected by real-time fluorescent quantitative-PCR and immunohistochemistry, respectively.The correlation between the expression of TIM-3 protein and the clinicopathological features of patients with ESCC was assessed using Pearson's χ2 test and Fisher's exact test.Kaplan-Meier method and log-rank test were used to compare the postoperative survival among ESCC patients with different expression levels of TIM-3 protein.
Results:The expression of TIM-3 mRNA was significantly higher in ESCC tissues than that in the corresponding para-cancerous normal tissues(P<0.001).The expression of TIM-3 protein was found in 85.9%(55/64) of ESCC tissues, while it was very weak or almost negative in the corresponding para-cancerous normal tissues(P<0.001).The abnormal expression of TIM-3 protein was closely correlated with the depth of invasion, lymph node metastasis and TNM stage(all P<0.05).Meanwhile, the expression level of TIM-3 protein was negatively correlated with postoperative prognosis of ESCC patients(P<0.05).
Conclusion:The abnormal expression of TIM-3 may be closely related to the occurrence and development of ESCC.Therefore, TIM-3 may have potential clinical value in early diagnosis, prediction of prognosis and targeted therapy of ESCC.
