摘 要
目的 研究DHA复合物的抗癌机理。方法 用流式细胞术研究了DHA复合物对荷瘤鼠H22细胞和T淋巴细胞细胞周期及凋亡的影响。结果 (1)与阴性对照组相比,DHA复合物中、高剂量组G0-G1期H22癌细胞百分比明显增加(P<0.01);DHA复合物低、中、高剂量组G0-G1期T细胞百分比明显减小(P<0.01)。(2)与阴性对照组相比,DHA复合物低、中、高剂量组S期H22癌细胞百分比明显减小(P<0.01),S期T细胞百分比明显增加(P<0.01)。(3)与阴性对照组相比,DHA复合物中剂量组G2-M期H22癌细胞百分比显著减少(P<0.01),DHA复合物低、高剂量组G2-M期H22癌细胞以及DHA复合物低、中、高剂量组G2-M期T细胞百分比均显著升高(P<0.01)。(4)与阴性对照组相比,DHA复合物低、中、高剂量组H22癌细胞增殖指数(PI)明显下降(P<0.01),T细胞增殖指数明显升高(P<0.01)。(5)与阴性对照组相比,DHA复合物低、中、高剂量组H22癌细胞及DHA复合物中、高剂量组T细胞凋亡率明显升高(P<0.01)。结论 DHA复合物可抑制H22癌细胞的增殖,促进T淋巴细胞增殖,同时DHA复合物还可促进H22细胞、T淋巴细胞凋亡














Abstract
Objective To research the anti tumor mechanism of DHA compound. Methods Flow cytometry was used to study the effect of DHA compound on cell cycle and apoptosis of H22 cell and T cell in mice bearing tumor. Results (1) Compared with control, the medium and high dose DHA compounds increased the percentage of G0-G1 phase H22 cell significantly(P<0.01); the low, medium and high dose DHA compounds reduced the percentage of G0-G1 phase of T cell obviously(P<0.01). (2) Compared with control, the percentages of S phase of H22 cell in three groups with DHA compound were lower significantly(P<0.01). But the percentages of S phase of T cell increased obviously(P<0.01). (3) Compared with control, the percentage of G2-M phase of H22 cell in DHA medium dose group was lower significantly(P<0.01).But the percentages of G2-M phase of H22 cell in the other two DHA groups and G2-M phase of T cell in all DHA compound groups were higher obviously(P<0.01).(4) Compared with control, in DHA groups, the PI of H22 cell decreased and that of T cell increased obviously(P<0.01).(5) Compared with control, the percentage of apoptosis of H22 cell increased obviously in all DHA groups(P<0.01),in medium and high dose DHA groups, the T cell apoptosis rates were higher (P<0.01).Conclusion DHA compound can inhibit H22 cell growth and promote T cell proliferation. At the same time, it can accelerate the apoptosis of both H22 cell and T cell.
