EGFRvⅢ与S100A9在人脑胶质瘤中的表达、相关性及其与预后的关系
         
Expressions and correlation of EGFRvⅢ and S100A9 in human glioma and their relationships with prognosis

摘    要
目的:探讨表皮生长因子受体变异体Ⅲ(epidermal growth factor receptor variant Ⅲ,EGFRvⅢ)和S100钙结合蛋白A9(S100 calcium binding protein A9,S100A9)在人脑胶质瘤组织中的表达水平及其相关性,以及2种蛋白表达与胶质瘤预后的关系。方法:收集重庆医科大学第二附属医院2008年1月—2015年4月收治的110例经病理确诊的新发胶质瘤标本及其临床资料。同时,收集10例非肿瘤脑组织作为对照。采用免疫组织化学法检测胶质瘤标本和对照脑组织中EGFRvⅢ和S100A9蛋白的表达水平,运用Spearman等级相关分析法分析蛋白表达与胶质瘤患者临床病理因素的相关性。随访其中75例胶质瘤患者,实访67例,采用 Kaplan-Meier法对随访资料进行生存分析。结果:低级别胶质瘤(Ⅰ~Ⅱ级)中EGFRvⅢ的中高表达率为20.00%,而高级别胶质瘤(Ⅲ~Ⅳ级)中EGFRvⅢ的中高表达率为38.00%,两者差异有统计学意义(P< 0.001);低级别胶质瘤(Ⅰ~Ⅱ级)中S100A9的高表达率也明显低于高级别胶质瘤(Ⅲ~Ⅳ级)中S100A9的高表达率(11.67% vs 66.00%,P< 0.001)。EGFRvⅢ和S100A9蛋白表达水平之间呈明显的正相关性(rs= 0.785,P< 0.001)。Kaplan-Meier生存分析表明,EGFRvⅢ和S100A9蛋白均表达阳性的胶质瘤患者的预后较差(P= 0.002)。结论:EGFRvⅢ和S100A9蛋白均随着胶质瘤恶性等级升高而表达增强。联合检测EGFRvⅢ和S100A9蛋白水平可以有效判断胶质瘤患者的恶性程度及预后,有助于为胶质瘤患者制定个体化的治疗方案。

Abstract
Objective: To investigate the expression levels of epidermal growth factor receptor variant Ⅲ (EGFRvⅢ) and S100 calcium binding protein A9 (S100A9) in human glioma tissues and their correlation, as well as the relationship between the two protein expressions with the prognosis of glioma. Methods: The pathological specimens and clinical data of 110 patients with pathologically confirmed primary glioma were collected from the Second Affiliated Hospital of Chongqing Medical University from January 2008 to April 2015. Besides, 10 cases of normal brain tissues were collected as the controls. Immunohistochemistry was used to detect the expression levels of EGFRvⅢ and S100A9 in glioma specimens. The Spearman rank correlation analysis was used to explore the correlation of the two protein expressions with the clinical and pathological factors of glioma patients. Seventy-five cases of glioma were followed-up, finally 67 cases finished the follow-up. Kaplan-Meier curve was drawn for survival analysis of follow-up data. Results: The middle- and high-expression rate of EGFRvⅢ in low-grade glioma was 20.00%, while it was 38.00% in high-grade glioma; there was significantly difference between them (P < 0.001). The high-expression rate of S100A9 in low-grade glioma was also lower than that in high-grade glioma (11.67% vs 66.00%, P < 0.001). In addition, there was significantly positive correlation between EGFRvⅢ and S100A9 protein expressions (rs = 0.785, P < 0.001). Kaplan-Meier survival analysis showed that the prognosis of patients with co-positive EGFRvⅢ and S100A9 was poor (P = 0.002). Conclusion: With the increase of malignant grade of glioma, the expression levels of EGFRvⅢ and S100A9 were also elevated. Combined detection of EGFRvⅢ and S100A9 proteins can be more accurate in evaluating the grade of malignancy and predicting the prognosis of glioma, hence, it is helpful to make individualized treatment programs for the patients with glioma.
